Here are some ideas.
Reinstall wow.
Run repair.exe
Open up ports in your firewall and router
Run as admin if you are not.

In case someone else reaches this paging searching google for:
"launcher cannot obtain patching information. please check your internet configuration"
What worked for me was turning off my anti-virus software for a moment.
I'm using Avira, had the same little annoying window, disabled Avira and started up again, worked like a charm.
After first launch, I simply re-activated Avira.

It happens when it needs to update and my anti virus protection is on. Turning it off till the patch is complete does the trick

Found a solution.
(for none proxy users)
Go to Internet Explorer -> Tools -> Internet Options -> Connections tab -> LAN Settings. Once you're there make sure only the "toggle the Automatically Detect Settings" check-box is checked and apply the setting. Try the Launcher or game again.

Totally bizarre, but aceman's tip worked for me as well. I was in game, then suddenly, disconnected. My wireless connection was fine since I could access all web sites (except, strangely, those associated with WoW). However, conducting this simple fix resolved my connection/launcher issue.

Brand New PC build with Windows 7.
Dunno what happened but after installing all my Blizzard games and played them the Launchers just failed to work after installing Skype, Yahoo! Messenger and a few other IM's. (don't judge!)
The Launchers wouldn't work as long as those programs were running i searched and searched the Internets. (loads of good info but no working solutions)
I did the Aceman's suggestion once(or twice) but never messed with it cause it was already checked and after seeing it again went back and looked at all the settings.
Since "Auto Detect" was already on i thought "Why Not" and unchecked it and left it off and the Launchers started.
Sometimes i think Windows is just all Derptastic and doesn't understand its basic functions and thinks a "Yes" really is a "No".
TL:DR - Test all options while in both "On" and "Off" state.
